Everytime I startup Lightwright (mac), I have to re-enter my shared folder location. I want it to stay on my onedrive so my labels and everything are available to each of my computers.

First I've heard of this. I'd start by making sure you are running the current version of Lightwright (6.0.13), available via the Downloads menu at mckernon.com.
The only reason I can see the shared folder location not being saved is if Lightwright doesn't have write privileges to it, or if it's not available when Lightwright launches.

I am running the latest Version of LW. On windows I know I could run program as administrator and I would gain access, however, I am fairly new to Mac and do not know the equivalent. While I am saving to Onedrive folders, these files and folders are locale to my machine, so I don't know how they wouldn't be available at the start of LW.

It's hard to say, I generally recommend not trying to share Lightwright files on Dropbox, because Dropbox may be doing a microsecond-long sync operation at the same moment that Lightwright tries to access the file, so the operating system tells Lightwright the file is busy at that moment. First, be sure you're setting a folder on OneDrive to be the shared location, not the whole OneDrive, then check to make sure that you have read/write permissions for the folder you want to set as the shared location. You do that by selecting it and choosing Get Info (Cmd+I). Sharing and Permissions are at the bottom of the Get Info window.
